Sword destroys a Horror that has been killing people, causing considerable destruction in the process. Meanwhile, Sophie consults the fortune teller "Moon Wizard" to locate her missing brother, but he turns into a Horror. She only just escapes with her life and then Sword arrives on his motorcycle and destroys it. Sword then takes her to his favorite diner and Makai headquarters for a meal. It appears that they both seek "El Dorado". He promises to protect her although he also asks her to be bait for a Horror who has been hunting red-haired women. The strategy works and Sword destroys the Horror, however again causing considerable destruction.

Jukai is a doctor who travels the land fitting prosthetic arms and legs to those who have lost their limbs in war. One day, on the riverbank he finds a baby missing most of its body parts. He decides to raise the child, who he names “Hyakkimaru”. Jukai provides the child with prostheses, but the time comes when Hyakkimaru sets out on a journey to recover his own body parts.

It all began ten years ago. D2s attacking Boston, a blood-soaked concert hall: The incident known as the Boston Tragedy. The conductor who lost his life in that tragedy was none other than Kenji Asahina, Takt's father. After losing his father, the boy decides to devote himself to playing the piano and he lets his rage play through his melodies. The one who took care of him was a girl named Cosette, who looked just like Destiny.

After entering Lord Daigo's domain, Dororo and Hyakkimaru hear tales of Banmon, the huge wall that is the last remnant of a fort that once stood on the border. Hyakkimaru squares off against the fox demon that haunts the wall.

Sabame has been feeding people to the demons to protect the village over which he rules. As Hyakkimaru once again confronts the demon moths, Dororo gets trapped in a storehouse.

Inside Daigo's castle, Lady Nui appears before the cell in which Dororo has been imprisoned. Meanwhile, Hyakkimaru is also approaching the castle as he tries to take back both his body and Dororo. Behind Hyakkimaru stands Midoro, who has transformed into a ghoul.

Takt and crew finally reach their destination of New York. They reunite with Charlotte, Anna's older sister, who works for the Symphonica's technological development department. Charlotte begins to examine Destiny. Who is an incomplete Musicart. But then, Anna realizes something is wrong with Takt. Daewi shows no mercy in his fight against Mira with the goal of getting the money to help his friend, Woo Seungtae, who suffers from an incurable disease. Mori handily defeats his own semifinal opponent, Brazilian jiu-jitsu user Byeon Jaehee, but is angered by Daewi’s method of defeating Mira by targeting her injuries. Daewi, meanwhile, learns that Seungtae has taken a turn for the worse and strengthens his resolution even further. Then, amidst all this, Mori and Daewi face each other in the final round.

Cosette transformed into the Musicart named Destiny. Destiny proclaims that her mission is to eliminate the D2s. Destiny bestows a conductor's baton upon Takt, and he uses his Conductor powers for the first time. And then, they are confronted by two individuals: Lenny and Titan. They are an official Conductor and Musicart from the New York Symphonica, which was the strategic headquarters to fight the D2s.

A giant Black Night Siderite starts to stir as Sagan hums a certain melody. The Black Night Siderite that grows as big as a tree starts to swallow up the New York Symphonica. Sagan has finally made his move to make his ambitions come true. As Takt and Destiny witness this eerie spectacle, they find out that Anna and Charlotte are still inside the Symphonica.

The god that appeared over the arena has been shot down by Jaesan's technique. Mori and the rest have fallen into a huge cave, where they battle against Jegal, who has transformed into a god after absorbing the Key. Mori, Daewi, and Mira face him together, but their attacks don't even reach him, and Daewi is severely wounded protecting Mori from Jegal's vicious onslaught. As the situation grows desperate, a memory that had remained dormant in Mori's mind all this time reawakens...

Dororo and Hyakkimaru come across an abandoned, burnt-out temple, where they meet a man named Sabame who rules the surrounding area. Sabame invites the two of them to his residence and tells them the story behind the burnt-out temple and the ghoul that haunts it. However, Dororo and Hyakkimaru suspect that he might not be telling the truth.

Takt and crew stop by a certain town. All of the young people had fled the town, leaving only the elderly. The residents happen to ask Anna and Destiny for help. Meanwhile, Takt wandered around town on his own and happens to hear a melody quietly resonating from an old bar. It was definitely the sound of music.

After learning that his grandfather, Jin Taejin, has been confined, Mori rushes to set him free. Daewi and Mira await his arrival at the tournament with no idea what's going on. With Mori missing and Daewi suspended, Mira is forced to kick off the team match alone. She struggles against her opponent, Marin, who evades her attacks and counters with her own sword. When he calls it a National Treasure, it begins to glow white, and its true form is revealed...

The national God of High School tournament will be a team battle between teams formed from the top three participants in each regional preliminary. Mira wins the battle for third place, meaning she will compete in the nationals with Mori and Daewi. Mujin explains to the other administrators that this is all part of the search for the one with the potential to be the "Key." At the same time, Mujin puts out a call to The Six, a group of extraordinarily powerful fighters, in order to deal with Nox, another organization searching for the Key.

Takt and Destiny learn how to fight properly from Lenny and Titan as they continue to make their way to New York. However, Takt and Destiny still have a hard time being in sync. They then coincidentally run into Lenny's old buddy Jonathan. Jonathan then leads them around his home, Las Vegas.

Ilpyo mercilessly beats down Jegal. Just as he's about to deal the finishing blow, Ilpyo sprouts tentacles and ensnares him. With the charyeok Greed, Jegal unleashes the charyeok powers of the others he's absorbed and turns their incredible powers upon Ilpyo. As their battle rages, something changes in the sky over the arena, and it is soon filled with a sword-wielding God and his many followers...

Four years had passed since Sagan's called a truce with the D2s. Music was still taboo throughout the world, and melodies ceased to be heard. Somewhere in America, which was now a wasteland, a little boy who grew up not knowing what music is hears the melody from a piano. The one playing it is a young man with black hair named Takt. And then, a deformed creature known as a D2 and a girl named Destiny, who is armed with a sword, appear as though they were lured in by the melody.
